چکیده

Cry4Aa toxin produced by Bacillus thuringiensis subsp. israelensis exhibits specific toxicity to larvae of medically important mosquito genera. In the present study, we analyzed characteristics channel pores formed recombinant in a solvent-free planar lipid bilayer. Stable currents were observed electrophysiologic measurements, and single-channel conductance was 187 ± 10 pS symmetrical buffer containing 150 mM KCl. The cation-selective, with an estimated PK/PCl permeability ratio 4.9. addition, exhibited apparent cation preference order Na+ > K+, Ca2+, K+ Ca2+. Although effect limited, seemed be correlated toxicity. Culex pipiens reared NaCl solution greater sensitivity Cry4Aa, particularly early period after exposure. presence cations that preferentially translocate through may facilitate excessive influx water into midgut cells, leading colloid-osmotic lysis. Whereas CaCl2 had some on mosquito-larvicidal activity KCl no effect. mitigated variety ion channels cell membrane.

Structure of the functional form of the mosquito larvicidal Cry4Aa toxin from Bacillus thuringiensis at a 2.8-angstrom resolution.

The Cry4Aa delta-endotoxin from Bacillus thuringiensis is toxic to larvae of Culex, Anopheles, and Aedes mosquitoes, which are vectors of important human tropical diseases. With the objective of designing modified toxins with improved potency that could be used as biopesticides, we determined the structure of this toxin in its functional form at a resolution of 2.8 angstroms. Like other Cry del...

Crystallization and preliminary crystallographic study of the functional form of the Bacillus thuringiensis mosquito-larvicidal Cry4Aa mutant toxin.

The 65 kDa functional form of the mosquito-larvicidal Cry4Aa-R235Q mutant toxin has been crystallized. The crystals belong to space group C222(1), with unit-cell parameters a = 91.2, b = 202.1, c = 98.7 A, and contain one molecule per asymmetric unit. The crystals diffract to approximately 2.9 A using synchrotron radiation and a complete native data set has been collected. The structure has bee...

Mosquito larvicidal activity of transgenic Anabaena PCC 7120 expressing toxin genes from Bacillus thuringiensis subsp. israelensis.

Genes encoding the mosquito larvicidal toxins Cry4Aa, Cry11Aa, Cyt1Aa and the regulatory P20 from Bacillus thuringiensis subsp. israelensis were introduced into the nitrogen-fixing, filamentous cyanobacterium Anabaena PCC 7120 for expression under control of two strong promoters P(psbA) and P(A1). The clone pRVE4-ADRC displayed toxicity against fourth-instar larvae of Aedes aegypti, the highest...

Analysis of mosquito larvicidal potential exhibited by vegetative cells of Bacillus thuringiensis subsp. israelensis.

Vegetative Bacillus thuringiensis subsp. israelensis cells (6 X 10(5)/ml) achieved 100% mortality of Aedes aegypti larvae within 24 h. This larvicidal potential was localized within the cells; the cell-free supernatants did not kill mosquito larvae. However, they did contain a heat-labile hemolysin which was immunologically distinct from the general cytolytic (hemolytic) factor released during ...

Structural Basis of Pore Formation by Mosquito-larvicidal Proteins from Bacillus thuringiensis

The insecticidal character of the three-domain Cry -endotoxins produced by Bacillus thuringiensis during sporulation is believed to be caused by their capability to generate lytic pores in the target larval midgut cell membranes. This review describes toxic mechanisms with emphasis on the structural basis of pore formation by two closely related dipteran-specific toxins, Cry4Aa and Cry4Ba, whic...
